Abstract
Thepharmacokineticsofdecursinanddecursinolangelate(D/DA)wasinvestigatedinmaleSD
ratsfollowingoralandintravenousadministration.D/DAandmetabolitesobtainedfrominvitro
sampleswereevaluatedbyLC/MS.ThelevelofD/DAandmetabolizeddecursinolintheblood
followingoralandintravenousadministrationdeclinedaccordingtofirstorderkinetics,withT1/2
valuesof56.67,58.01and57.22h,respectively,beingobservedafteradministrationofadoseof
2mg/kgbodyweight.Thelargeintestinewasthemajorsiteofdispositionfollowingoral
administration.ThesedataindicatethatD/DAisrapidlyabsorbedfromthegastrointestinaltract.
Ininvitroexperimentutilizinglivermicrosomalprotein,themajormetabolicreactionofD/DA
occurredtochangedecursinol.Thecumulativebiliary,urinary,andfecalexcretionofD/DAinbile
ductcannulatedratswas36.10+/2.9,25.35+/3.8,and34.20+/3.2%,respectively,at72hafter
administration.TheseresultsindicatethattheabsorptionofD/DAisalmostcomplete,andthatits
metabolitesareprimarilyexcretedintofecesthroughthebile.TheseresultsindicatethatD/DAis
subjecttoenterohepaticcirculation.
