MS Interleukin 6 merupakan sitokin inflamasi yang diproduksi oleh bermacammacam sel limfoid dan sel nonlimfoid seperti sel T, sel B, monosit, fibroblas, keratinosit, sel endotel, sel mesangium, beberapa sel tumor dan jaringan lemak. Kadar interleukin 6 meningkat pada binatang dan manusia dengan kadar glukosa darah yang tinggi sehingga meningkatkan resiko terjadinya diabetes melitus . Ekstrak daun pare mengandung charantin, polypeptide-P dan vicine yang memiliki efek seperti insulin. Penelitian ini bertujuan untuk mengetahui efek ekstrak daun pare (Mamordica charantia) terhadap kadar interleukin 6 (IL-6) serum tikus(Rattus novergicus) Wistar model diabetes melitus dengan injeksi streptozotocin dan diet tinggi lemak. Penelitian ini menggunakan studi true experimental, dilakukan pada 25 ekor tikus Wistar jantan yang dibagi secara acak menjadi 5 kelompok. Kelompok 1 adalah tikus diberi diet normal saja selama 10 minggu (kontrol negatif), kelompok 2 tikus diberi diet tinggi lemak saja dan injeksi streptozotocin (STZ) (kontrol positif), sedangkan kelompok 3 sampai dengan 5 (3 kelompok) diberi diet tinggi lemak selama 35 hari dan injeksi STZ, kemudian diberi asupan ekstrak daun pare dengan dosis berbeda (42, 75 dan 133mg/100grBB) secara per oral dengan sonde setiap hari sekali selama 14 hari. Selama pemberian ekstrak daun pare, diet ketiga kelompok (3 sampai dengan 5) tetap diberikan diet tinggi lemak. Parameter yang diukur adalah interleukin 6. Analisis data menggunakan metode One Way ANOVA dilanjutkan uji Post Hoc Tukey, uji regresi dan uji korelasi. Analisa data menunjukan bahwa pemberian ekstrak daun pare berpengaruh terhadap kadar interleukin 6 secara signifikan (p<0,05) dengan mode regresi Y= 1338,296 - 4,483X dan memiliki hubungan cukup kuat (r>0,5). Kesimpulan penelitian ini adalah ekstrak daun pare (Mamordica charantia) mampu menurunkan kadar interleukin 6 serum tikus (Rattus novergicus) strain Wistar model diabetes melitus dengan injeksi STZ dan diet tinggi lemak. Interleukin 6 is an inflammatory cytokine produced by both lymphoid cells and non lymphoid cells such as, T cells, B cells, monocyte, fibroblast, keratinocyte, endothelial cells, mesangium cells, several tumor cells and adipose tissue. The level of interleukin 6 increases in animal and human with high blood glucose, resulting in an increase of the risk of diabetes mellitus. Bitter melon leaves extract contains charantin, polypeptide-P and vicine which have insulin-like activity. This aim of this experiment is to determine the effect of Mamordica charantia leaves extract on serum interlukin 6 (IL-6) Level of Wistar rats (Rattus novergicus) model diabetes mellitus injected by streptozotocin (STZ) and high fat diet. This experiment used true experimental, done on 25 male Wistar rats, that were randomly divided into five groups. Group 1 was rats fed by standard diet for 10 weeks (negative control), group 2 was rats fed high fat diet and injected by streptozotocin (STZ) (positive control), group 3 to 5 (3 groups) were rats fed by high fat diet for 35 days and injected by STZ, then were given different doses of bitter melon leaves extract (42, 75, and 133mg/100grBB) per oral per day for 14 days. During bitter melon leaves extract treatment, group 3 to 5 rats still fed by high fat diet. Parameter that was measured was interleukin 6 level. Data analysis which used the One-Way ANOVA method followed by Post Hoc Tukey Test, regression test and correlation test. Data analysis showed that bitter melon leaves extract had effect on interleukin 6 level of Wistar rats significantly (p<0,05), with regression mode Y= 1338,296 - 4,483X and had a strong enough correlation (r>0,5). The conclusion of this experiment is bitter melon leaves extract can decrease serum interleukin 6 level of Wistar rats model diabetes mellitus injected by STZ dan high fat diet.
