Pengaruh Pemberian Monosodium Glutamate (MSG) terhadap Gambaran Histologis Testis Mencit

Kurniawan Adi Putranto1, Endang Listyaningsih S.2, Jarot Subandono3 Tujuan Penelitian: Melihat perubahan struktur histologis testis pada mencit yang diberi Monosodium Glutamate (MSG) dalam dosis rerata konsumsi di Indonesia dan mengetahui hubungan peningkatan dosis Monosodium Glutamate (MSG) dengan penurunan jumah spermatid. Metode Penelitian: Eksperimental laboratorium dengan the posttest only control group design. Mencit jantan sebanyak 27 ekor dibagi dalam 3 kelompok yaitu kelompok kontrol (K) dan kelompok perlakuan 1-2 (PI-PII). Kelompok kontrol mencit diberi aquades selama 10 hari berturut-turut. Kelompok perlakuan I (PI) diberikan Monosodium Glutamate 1,56 mg/20 g BB per hari melalui oral selama 10 hari. Kelompok perlakuan II (PII) diberikan Monosodium Glutamate 3,12 mg/20 g BB per hari melalui oral selama 10 hari. Pada hari ke-11, mencit dikorbankan dan diambil testisnya untuk pembuatan preparat dengan pengecatan HE kemudian diamati dengan menghitung jumlah sel spermatid pada preparat testis kanan dan kiri dalam satu kelompok mencit. Data dianalisis dengan uji Oneway Anova dan uji LSD. Hasil Penelitian: Jumlah rerata sel spermatid kiri dan kanan pada kelompok kontrol adalah 126,56 17,42; kelompok perlakuan I (PI) adalah 74,22 10,46; dan kelompok perlakuan II (PII) adalah 51,78 6,64. Hasil uji statistik Oneway Anova menunjukkan adanya perbedaan yang bermakna dalam ketiga kelompok penelitian (p = 0,00). Hasil uji statistik LSD juga menunjukkan perbedaan yang bermakna antara ketiga kelompok dengan masing-masing p = 0,00. Simpulan Penelitian: Pemberian Monosodium Glutamate (MSG) dalam dosis konsumsi rerata di Indonesia dapat mempengaruhi gambaran histologis testis mencit yang ditandai dengan penurunan jumlah sel spermatid mencit. Jumlah sel spermatid semakin menurun seiring dengan semakin besarnya dosis Monosodium Glutamate (MSG). Kata Kunci: monosodium glutamate, dosis konsumsi Indonesia, testis

The Effect of Monosodium Glutamate (MSG) Usage on Histological Structure of Mices Testis

Kurniawan Adi Putranto1, Endang Listyaningsih S.2, Jarot Subandono3 Objective: This study aims to look at changes in histological structure of testis in mice fed Monosodium Glutamate (MSG) in a dose of average consumption in Indonesia and also to test the corelation between Monosodium Glutamate (MSG) dose increase to spermatid cells decrease in amount. Methods: Experimental laboratory with th posttest only control group design. Twenty-seven male mice were divided into 3 groups: control group (K) and treatment group 1-2 (PI-PII). The control group mice were given distilled water for 10 consecutive days. The treatment group I (PI) being tested per day for 10 consecutive days by giving Monosodium Glutamate for 1,56 mg/20 g of body weight. The treatment group II (PII) were given Monosodium Glutamate 3,12 mg/20 g of body weight per day orally for 10 days. On day 11, mice were killed and testis were taken for making preparations with HE painting. Histological structure of mices testis was observed by counting the number of spermatid cells on the right and left testicular preparations in one group of mice. The data analyzed by Oneway Anova test and LSD. Results: The average number of left and right spermatids cells in the control group was 126.56 17.42; treatment group I (PI) was 74.22 10.46; and treatment group II (PII) was 51.78 6.64. Oneway Anova statistical test results indicate a significant difference in all three study groups (p = 0.00). LSD statistical test results also showed significant differences between the three groups in comparison p = 0.00. Conclusion: The effect of Monosodium Glutamate (MSG) in a dose of average consumption in Indonesia changes histological structure of testis which the sign is the decrease in the number of spermatid cells on mice. The number of spermatid cells decreased along with the increased of given doses of Monosodium Glutamate (MSG). Keywords: monosodium glutamate, consumption dose in Indonesia, testis
