Procedure for Spray Test of Roundup Ready Canola
Updated Feb 2009

MATERIALS

Equipment: Knapsack Sprayer 15L
Nozzle 80 02 standard flat fan
Constant Flow Valve 3 Bar

Chemical: Roundup Ready Herbicide (Monsanto) (granular)
Active constituent: 690g/kg GLYPHOSATE (present as mono-ammonium salt)

Potting Mix: Available from research bunker

Seed: Representative sample of line being tested, a susceptible line (ie.
susceptible control) and a tolerant line that has been tested and passed (ie.
tolerant control)
Trays: 2 x 100 cell grow trays (for variety to be tested)
2 x 100 cell grow tray (1 for susceptible control, 1 for tested & passed line)

Spray rate for chemical: 0.9kg product per hectare
Chemical required using equipment as noted:
Mix 1.6g RuR Herbicide/1 Litre Water
Knapsack: Pump to maximum pressure


METHOD
Using potting mix, fill all trays.
Sow a number of seeds from each line into each cell of relevant trays, water lightly and
grow using normal horticulture practices.

When seedlings appear, thin cells, allowing for attrition i.e. keep a number of extra plants
above 100.
Before spraying remove extra plants, leaving 100 plants per tray.

Plants should be sprayed at full cotyledon to two leaf stage.

Align trays to be sprayed lengthwise in suitable area for spraying.
Hold spraying wand at height to produce a band width of 32cm.
Spray trays walking at medium pace with one sweep.
Label trays : Variety, batch, grower, date planted, date sprayed and name of test (RuR)

ASSESSMENT

When control variety plants are dead (approx 10-20 days after spraying), conduct a count
of tolerant and susceptible plants of variety being tested.

Record results in QC database and in RR testing summary spreadsheet.

If results obtained from the spray test differ from what is expected, note it in the QC
database and refer to the Genetic Purity Manager immediately to discuss further action.
