1. Wash your hands.





2. Dry them with a clean towel or tissue.
3. Read the medication label.
4. Shake the bottle as this is indicated on the bottle or it is directed by the doctor.
5. Unscrew the cap of the bottle (for bottles with an integrated dropper, draw some liquid into the
dropper).
6. Lie down or tilt your head back and gently pull your lower eyelid down.




7. Hold the dropper (tip down) with the other hand, as close to the eye as possible without
touching it



8. While looking up, gently squeeze the dropper so that a single drop falls into the pocket made by the l
lower eyelid. Remove your index finger from the lower eyelid



9. Close your eyes slowly and gently press a finger against the inside corner of each eye and keep them
closed for approximately 1 minute to allow the medication to absorb. Straighten your head and wipe
away any extra liquid.

10. If you have more than one drop of medicine to put in your eyes, wait about 5 minutes after the first
medicine.

5. Carefully, place a line (1cm) of ointment inside the lower lid while being careful not to touch your eye
with the tip of the tube.





6. Your vision can become blurry for a few moments, but it will clear quickly.
7. Close you eyes slowly and gently press a finger against the inside corner of each eye and keep them
closed for approximately 1 minute to allow the medication to absorb.




8. Straighten your head and wipe away any extra ointment.

If you are prescribed
both eye drop and eye
ointment together use
the eye drop first.

5. Lie on your side or tilt your head over so that the ear which needs the drops is facing upwards.
6. Pull your earlobe upwards (or if giving to a child younger than 3 years of age, pull backward and
downward) to open the ear canal and squeeze/drop the correct number of drops into the ear.





7. Keep your head tilted for about five minutes so that the drops can spread into the ear.





8. Straighten your head and wipe away any extra liquid.
9. Replace the cap on the bottle.

7. Hold the bottle or dropper above your nostrils and gently squeeze the correct number of drops into the
nostrils.

Squalamine Eye Drops
What is Squalamine?
Squalamine is an anti-angiogenic amino sterol derived from cartilage
of the dogfish shark, Squalus acanthus..
Mechanism of Action
It blocks ion transporters that regulate cell function by controlling
pH and metabolism.
When bound to calmodulin, squalamine also blocks the action of
vascular endothelial growth factor (VEGF) and integrin
expression,thereby inhibiting angiogenesis (stop formation of new
blood vessel) in the retina and macula.
A History of Squalamine
Research
Initially, Genaera Corporation
developed squalamine as an intravenous formulation for wet-age
related macular degeneration (AMD) a degenerative retinal eye disease that
causes a progressive, irreversible, severe loss of central vision.
After Phase 1 and Phase 2 clinical trials, this project was discontinued due to
financial and subject-recruitment issues in 2007.
Squalamine has been shown to inhibit new vessel formation without significant regression of existing
lesions, and, depending on the lesion composition, combination with adjunctive
therapies may be appropriate.
Then this project was acquired by Ohr Pharmaceutical, which continued the
development of squalamine in a topical eye drop formulation.
This study has entered phase II clinical trial to evaluate the safety and efficacy of topical ophthalmic
squalamine lactate eye drops and expected to complete in December 2014.

Current Treatment
VEGF inhibition therapy is the best available treatment for wet AMD but is limited by the need for
repeated intravitreal injections.
Options currently include either a full-length recombinant monoclonal antibody (bevacizumab/
Avastin) or a highly afnitized fragment of the antibody (ranibizumab/Lucentis), or a pegylated
aptamer (pegabtanib sodium/Macugen).





Conclusion
Thus, with the development of squalamine eye drop which designed for self administration, may
provide several potential advantages over the FDA approved current standards of care, which
require intravitreal injections directly into the eye.
