~

Did you hear about the elderly woman who was pulled over
for speeding? She explained to the young officer, I had to
hurry up before I forgot where I was going.

Memory lapses like these can produce some amusing


situations. We can laugh knowing that often senior
moments are fairly harmless. However, mild memory
problems can also have consequences that negatively affect
our lives.

The Brain Health Guide is a tool to help keep your brain as


sharp as possible so that any memory lapses that do occur
are the laughable kind.

America's State of Brain Health

here is good news and bad news when it comes to brain health.
The good news is that people are living longer than any other
time in history. Major nutritional and medical advancements have
been made to help keep the body physically healthy. The bad news is
we are seeing an unprecedented number of mild brain health issues
affecting our families and our friends. When life spans were shorter,
people simply were not living long enough to see the effects of natural
brain aging that are
common today.
Scientists estimate
we lose between
30,000 and 50,000
brain cells every
single day. This leads
not only to common
occurrences like the
misplacement
of
car keys and reading
glasses, but difficulty with management of our time and household
tasks. A recent study by the Natural Marketing Institute ranked
healthy mental function as the number one health concern among
those over 60 years old.
As we age, mild memory problems result in more than difficulty
remembering. It also leads to an inability to focus, pay attention
or stay on task. With increasing age comes increasing stress that
can affect the brain. The health of the brain is arguably the most
important part of overall health because it controls all organ systems
in the body.
The problems with mild memory problems in the United States are
mainly due to the aging of our society. In the year 1900, the average

life expectancy was 47 years. In the year 2012, life expectancy


has risen to nearly 80 years. While this increase reflects positive
improvements in health care, it has led to an increase in the wear
and tear on our bodies, including our brains.
Caring for the brain requires an understanding of its needs. The
brain is a demanding organ. Despite comprising just 2% of the
average adult body weight, the brain requires 20% of the bodys
energy and up to 25% of the hearts blood flow. In each of the
100 billion neurons in the brain are energy factories known as the
mitochondria; the neuronal mitochondria turn glucose into chemical
energy in the form of adenosine triphosphate (ATP). Feeding your
brain the right nutrients becomes more important as we age.

Nutrition Tips for Memory Fitness

f were honest with ourselves, when we self-assess our overall


health, the brain is not usually considered. The parts of the body
giving us the most feedback get the most attention. Because of our
improving longevity, this is a major oversight, as the brain controls
everything the body does. We all know the brain controls cognitive
areas like memory, thinking, and focus. When these functions are
strongly supported, the implications on our lives are tremendous!
Julian Allan, lead researcher and health psychologist, concluded,
A person with less efficient executive function is less likely to resist
temptation and stick with what they had planned on any given day,
than someone with excellent executive function.

more later in the book but suffice to
say here that a properly supported
brain through proper nutrition,
lifestyle, and supplementation will
lead to a healthier more productive
life!
Unfortunately, the normal process
of aging can negatively affect brain
performance. The next several pages will give you valuable tips to
improve not only your executive function but the overall health of
your brain.
Tip #1 - Eat The Right Kind Of Fat:

Research suggests that a healthy diet can help keep your major
organs functioning at a high level. A diet consisting of the right
blend of proteins and complex carbohydrates has proven to be the
most beneficial. The brain and nervous system are comprised of
60% fat, so ensure your diet is rich in the Omega 3 essential fatty

acids found in cold water fish, fish


oil and flax oil.
Foods high in saturated fat, such as
fried and overly processed foods,
not only increase body mass but can
disrupt the natural function of the
brain. Poor diet can also affect your
cholesterol levels and blood pressure.
People with high cholesterol and
blood pressure are several more
times likely to experience age-related
mild memory problems. A good rule of thumb is skip the chips!
Tip #2 - Water, Water:

Heres an anti-aging tip so obvious we shouldnt have to mention


it: Water. Were bringing it up
because water doesnt get the credit
it deserves.
Used properly, water helps us look,
feel and act younger than we are
keeping healthy and enjoying life
to the fullest. If youre not already
doing it, drinking 8-12 eight ounce
glasses per day is the guideline. If
you prefer it bottled, four per day
would do the trick.
But dont cheat. Tea, coffee and soft drinks dont count. Substituting
these for water will actually dehydrate yousomething you dont need
and dont want.
Since it has zero calories, drinking water helps you lose weight and
keeps skin looking healthier. It keeps you more productive, regulates
body temps and helps in digestion. In general, consuming enough
water helps keep you in a good mood.
If you want to feel better, knock back at least 8 glasses of water a day.
You cant live without it, so drink up!

Tip #3 - Eat Your Vegetables:

New research has found that to retain the health benefits associated
with eating broccoli or other cruciferous veggies (like cabbage,
turnips, cauliflower, kale and broccoli), you must consume the real
thingbecause of a phytochemical inside them thats missing in
supplements.
The study, published by the Pauling
Institute at Oregon State University and
supported by the National Institutes of
Health, is one of the first of its type
to determine whether the healthy
compounds found in cruciferous
vegetables can be just as easily obtained
through supplements.
The answer is no.
You need the whole foods and must go easy on cooking them. While
some nutrients, like folic acid, can be better absorbed via supplement,
the compounds that give vegetables like broccoli their health value
comes from the real thing.
The reason is that an enzyme that must be there to provide the
maximum nutrient kick is absent in supplements of this type.
Without these enzymes, the absorption rate is five to eight times less.
Intensive cooking also impedes delivery. If broccoli is cooked until
it's mushy, its health value plummets.
Instead, cook for 2-3 minutes, or steam until it's still a little crunchy.
(Broccoli was highlighted because it contains the highest levels of a
class of phytochemicals that may reduce the risk of prostate, breast,
lung and colorectal cancer.)
Bottom line: Eat your vegetables! And consume the real thing, prepared
correctly.
Tip #4 - Try Coconut Oil:

Coconut oil has been consumed for thousands of years in tropical


cultures. However, its health benefits have only recently been

acknowledged in the United States.


Coconut oil provides a variety of health
benefits. Coconut oil can make your skin
and hair healthier, with many people
reporting that it has even made their
wrinkles disappear! Coconut oil also
has benefits for the brain. People who
consume coconut oil daily report that
it helps with their short-term memory
and allows them to think clearly, stay
focused, and have better concentration. Researchers believe this
most likely comes from the medium chain triglycerides in coconut
oil which are converted by the liver into fat called ketones. Ketones
are a great source of energy for brain cells.
Coconut oil users love that it is so easy to useyou can cook and
bake with it, spread it on toast, eat it plain, and even smooth it on
your skin. Make sure the coconut oil you buy is non-hydrogenated
and contains no trans-fat.
Tip #5 Balance your Diet and Supplements:

Eating a well-balanced diet is the best way to get the important


nutrients our bodies and brains need. Sometimes it is hard for
people to get enough of the nutrients they need through diet alone,
so supplements can help to ensure the right balance is achieved. But
supplements should not be used to replace a healthy diet.
An article appeared recently in a national magazine, about a
writer who after realizing that the taking of vitamin supplements
was something hed been missing out on, went on a self-imposed
five month regimen as recommended by a respected supplement
manufacturerand reported his experience.
This writer was about 30 years old and had been doing a good job of
taking care of himselfeating right and getting the proper amount
of exercisebut was lacking in supplement savvy.

Soon after beginning (and logging) his daily dosage, he began to feel
much better, and found himself waking up feeling more vigorous,
armed with a greater confidence in facing the challenges ahead.
Supported by the supplements, and without realizing it fully, he
began changing his eating habitsgoing off his low in calorie/
high in protein diet to one including junk food, causing an 8-pound
weight gain over the five month test.
At the end of his singular study, the physicians who examined him
said that his vitamin-induced euphoria and subsequent straying from
making good lifestyle choices was a form of compensation, but poor
excuse for falling off the food wagon. And their advice?
Dont do that.
Like every other life plan, moderation and balance should be the key,
and while its okay to over-indulge now and then, staying on course,
eating right and exercising to fit your age and general health, then
backing things up with a realistic daily dosage of vitamins, minerals,
(and supplements) is the prescription for success.
Tip #6 Practice Moderation

Everybody likes a salty snack or a sweet treat. It is hard to say no


to ice cream on a hot day or chips and dip while watching the big
game.
But just as excess salt and diets high in sugar are bad for our bodies,
eating too much of these can also be harmful for our brains. Many
people have experienced the crash after a sugar buzz, which can
leave them feeling tired, sluggish, and not able to think clearly. The
long-term effects of consuming too much sugar could leave you
feeling like you do after a sugar buzz all the time.
The best way to approach food items like French fries and cookies
are to eat them in moderation. Once in a while they are a great treat,
but eating them on a regular basis not only can cause harm to our
bodies and brains, but it can take the enjoyment out of indulging
every once in a while.
Anyone who has ever been on a diet can attest to the fact that
nutritional changes can be hard to stick to long-term. But a healthy

diet is important when it comes to brain health. Of course, change


doesnt happen overnight. If youre not following the nutrition tips
for better brain health already, try incorporating them one at a time
over the course of a few months. You may be surprised at the positive
changes you see!

Lifestyle Tips for Memory Fitness

n Chapter 2 we gave you a few nutritional tips to follow to


improve your brain health and memory. Maintaining a healthy
diet is an important part of brain health, but it isnt the only way
to better memory. There are also many lifestyle changes you can to
incorporate into your routine.

Tip #1 - Physical Exercise:

A great way to improve the health of


your entire body, including your brain, is
through physical exercise. Committing to
a regular form of exercise is one of the
most difficult things to do. It requires a
time commitment and dedication. The
good news is that an hour in a gym every
day is not a requirement for good health.
Consistent, low stress workouts, like a brisk
walk a few times a week or yoga, can be enough. Even everyday activities
like gardening and cleaning promote good brain health. The normal
process of aging can put stress on your brain, but regular exercise can
help by maintaining fine motor skills, improving concentration skills,
and support mental health.

Tip #2 - Mental Exercise:

Just as proper diet and physical exercise


are important, so is directly stimulating
your brain with mental exercises. Activities
such as crossword puzzles, word games and
math problems are not only fun, but also
strengthen connections within your brain.
If you continue to challenge your brain,
you may be able to curb mild memory
problems, and become a better problem

solver. Increasing scientific evidence is showing that you can fight aging
by challenging your mind.

Tip #3 - Embrace Technology

Many new technologies exist today that


allow people to communicate with a
variety of people instantaneously. By
using these new communication tools
in the right way, networks of people can
learn to collaborate globally, discussing
ideas and arriving at possible solutions.
You might seize on a problem, learn about it, and find what others are
thinking. Many decry that Facebook, Twitter, Linkedin and others are
a waste of time, keeping us from communicating properly with each
other. But maybe they deserve a deeper look into whats possible if we
use these new technologies to better ourselves and our world. Strange
as it may seem, indulging yourself in these possibilities builds a sense of
communityone that goes well beyond the neighborhood.

Tip #4 Balance Technology with


Real Life Interactions
While many, regardless of age, use todays
communications tools to share information
or to be aware of where theyre supposed
to be next Thursday at 9:00some have
made social networking a way of life.

And while much of this new order in our


lives is good, research is showing many are
spending too much of their time making use of the sophisticated tools
at their disposal. And a significant percentage is also attempting to work
more than one program at a time, an effort that can be both seen as
both negative and positive.
Multi-tasking involves that part of the brain that governs social and
reasoning behavior and could be detrimental to cognitive reasoning.
Visual and motor skills might be improved, but the loss of the ability to
focus and think things through in an orderly way is also possible.

One way to use social technologies such as facebook to our advantage


is to use it as a planning tool. Join a local group and attend an event, or
schedule a get together with family and friends. Interacting with people
face-to-face is another great way to keep your brain in top shape.

Tip #5 Engage with People

Actively engaging with others requires using different regions of


the brain. The unpredictability of conversation requires you to pay
attention in order to follow along and contribute to a dialogue. This is
called active listening. The opposite, passive listening, offers little
help for brain development. Socializing has a positive effect on the brain
because of the mental exercise involved. Research has shown having
a limited social network is associated with many health problems.
Scientists do not completely understand how social interactions improve
mental function. One theory is our social networks help manage stress
by allowing us to share our struggles and deal with stress, and giving us
a sense of purpose and well-being.

Tip #6 - Change your Routine and Try Something New

Stimulate your sensory side by breaking routine; shave or brush your


teeth with your non-dominant hand, take a shower and/or get dressed
with your eyes closed, and learn new skills. This could include taking
a course on a subject about which you know little, learning a new
language or preparing a dish from unfamiliar recipes. The key is to
choose something that really interests you.
In addition to challenging your brain with new experiences, here are
some memory improvement ideas you might try on sometime: Pay
attention. Focus on something for 8-10 seconds and process it in
your memory. Apply your best learning style. Do you pick up on
things best by reading, or by listening? Go with your strength. Involve
your senses. Relate things to color, texture, taste and smell if you can.
Organize information. Write things down, take notes regarding
complex material, then organize and study them. Interpret complex
material. Focus on the basics rather than memorizing myriad details.
Be able to explain it to someone else in your own words.
Finally, Be Motivated and Positive. Declare to yourself that you not

only want to remember what youve learned, youre able to do both.

Tip #7 Find Something to Love

Recent research on the phenomenon


of aging has found that having a stable
relationship and clearly defined goals
in life have as much to do with growing
older as an active, vibrant person as being
blessed with good genes.
If you take the steps to care for your
physical self by eating the right foods,
exercising regularly, getting enough sleep
the critical factor remaining is lovewhich encompasses not only your
feelings about the living beings around you, but the great things you can
do in your lifelike being persistent, curious, having a sense of humor,
and caring about what you do.
Having love in your life is in itself a source of energyhelping provide
reason, purpose, and direction. Generating, building, and maintaining
social connections are not only of immense help mentally, possessing
these traits bring physical support too. Having these attitudes percolating
from within not only brightens outlook, they strengthen overall wellbeing. Not all of us are lucky enough to be equipped with the full array
of genetic longevity, excellent physical and mental health, and lots of
love in our lives, but we can still do best with what we have. Maximizing
the efforts over that in which you have control gives you the best shot of
coming out on top. Then when you face life head-on, youll be making
the most of its positives and dealing firmly with its challenges.
It is important to recognize you can play an active role in the health
of your brain. Scientists have demonstrated that connections between
brain cells are constantly changing. A lack of attention to brain activities
will cause disconnections, while active care of the brain will promote
greater mental acuity.

Jellyfish & the Nobel Prize...
What's the Connection?

n addition to nutrition and lifestyle changes to support your brain


health, the addition of key supplements can play a key role in
addressing specific functions in the brain. Here we will explain how
the jellyfish and the Nobel prize played a role in the discovery of a
new tool that has researchers, physicians,
and consumers excited about the
prospects for better brain health in aging.
In 1961 a group of researchers from
Princeton University studied a specific
species of jellyfish called Aequorea
victoria to determine what caused them
to glow. Led by Osamu Shimomura,
PhD, they packed up their Plymouth
station wagon and drove to Friday
Harbor on San Juan Island near
Seattle, Washington. A constant stream
of glowing jellyfish floated by their
laboratory each day. They developed
an extraction process and eventually
obtained enough luminescent fluid for
study. The researchers discovered the cause of the glowing jellyfish
was a specific protein. Upon binding with calcium, this protein
triggered a small amount of light. Initially, this was a great discovery
of a novel aspect to this species of jellyfish called Aequorea victoria.
Later, a technique to manufacture this protein without harvesting
jellyfish was discovered, and a research purpose evolved from the
jellyfish protein.

Researchers began using the protein in experiments. Under a


microscope, they could track where calcium traveled in the body
because of the glow that occurred when this protein bound to
calcium--a biological flashlight! This jellyfish protein allowed
researchers to better understand the function of calcium in the body.
The pioneering discovery and the decades of research that followed
led to a 2008 Nobel Prize in chemistry for Drs. Shimomura, Martin
Chalfie, and Roger Y. Tsien.
Research conducted at the University of Wisconsin-Milwaukee in the
early 2000s helped determine that the jellyfish protein, apoaequorin,
could play a useful role in supporting brain performance in humans.
This discovery resulted in the formation of Quincy Bioscience, a
biotechnology company in Madison, Wisconsin, that develops
products and technologies based on apoaequorin to help support

in Madison, Wisconsin, and has been put through rigorous
scientific testing. In this chapter, a concise summary of this research
will be discussed.

It is clear when looking at the aging demographics in the United


States that supporting brain cell function needs to be a top priority!
Beginning January 1, 2011, the oldest members of the baby boomer
generation turned 65 years old. Since that day, and every day for the
next 19 years, 10,000 baby boomers will celebrate their 65th birthday.
Gray hair may look distinguished, but other consequences of aging
may not be as appreciated, such as mild memory problems.*
The supporting research behind Prevagen suggests that those who
are experiencing the beginnings of normal age-related mild memory
problems, as well as those who are not yet affected by memory lapses,

build on the strong evidence that had
been gathered on ability of apoaequorin
to improve memory.* The Madison
Memory Study was launched with
an ambitious goal to recruit over 200
people into a computer-based trial. The

concerns but be undiagnosed with any type of memory disorder. For


the next year-and-a-half, participants recruited for the study were
evaluated using a set of timed computer tasks.
The goal of the Madison Memory Study was to measure Prevagens
ability to improve brain function using computer software in people
experiencing normal age-related mild memory difficulties.*
Cogstate, Ltd., the worlds leading cognitive testing software
company, provided the testing protocol, which is the specific plan
for how the study was to be conducted. Computers were used in this
setting to objectively measure changes in cognition.

In 90 days, the Prevagen group significantly reduced the number of errors that were
made compared to placebo in the Groton Maze. This task measures executive function,
learning and memory.

The trial enrolled participants assigned to either Prevagen or placebo


for ninety days. Neither the people in the study, nor the researchers,
knew which capsule the participants were taking. This is known as
a randomized, double-blind, placebo-controlled study--the gold
standard for measuring the effect of a compound.
A total of 218 adults aged 40 to 91 years, were tested at predetermined
one-month time intervals and changes on specific assessments of
cognitive function were measured at various time points during the
study. The final results of the study were very encouraging. The
data showed that people on Prevagen had statistically significant
improvement in several areas of memory compared to baseline and
to placebo.*
The Prevagen group improved their scores in executive function,
learning, memory, and word recall.*

Short-Term Memory

Quick questionwhat is on your mental to do list, right now? Can


you recall them all?
The One Card Learning Task, is a part of the Madison Memory
Study that measures a participants ability to identify and compile
information on short-term memory.
The simplest definition of short-term memory is the state of being
currently aware of something and creating an action around it.
Like making and holding in your mind a small handful of to dos.
Or preparing a short list of prioritized tasksand remembering
to refer to it as you work through your day. Short-term memory is
used when meeting someone new, while reading a book, and during
conversation.
A fleeting thing, short-term memories are information that exits
quickly from your mind unless you make an attempt to keep it on

file. If you repeat an action, youre storing (or re-storing) it mentally,


providing greater meaning and association.
Short-term memory is just that, short. The average time a thought
can be retained is 30 seconds, but can be less. And the number of
bits and pieces we can hold in our brain varies. Most scientists agree
that 7 is a decent average number.
The One Card Learning task, sort of a sophisticated version of
the childhood card game Concentration, observes and analyzes a
subjects short-term memory skills. Participants taking Prevagen
scored highestanother indication it was supporting brain function.
From making your way through that small list of daily responsibilities,
to communicating effectively, short-term memory is a lifestrengthener.

Executive Function

Have you ever made a plan? You used your Executive Function,
which loosely means your brains ability to put an order to the things
in your world. You use it to do things like setting goals and priorities,
and identifying what to accomplish during our day.
Executive function is an extremely important function of the brain.
Should it falter, tasks become more difficult.
One of the tests in the Madison Memory Study is the Groton Maze
Learning (GML) task, used to measure participants executive
function. Looking at a tile-like grid on a computer screen, subjects
repeatedly learn to work their way through a maze that takes them
from an identified start to a marked ending. Errors are expected, but
if executive function is present, each time its taken, mistakes are
fewer. Those on Prevagen showed an almost double decrease in
errors over a specific time than those on the placebo.

a normal problem associated with aging. Results from the Madison

Delayed Recall

We exercise the function of delayed recall constantly. Have you ever


thought of something you wanted to accomplish at a later point in

Simple as it sounds, the ability to recall is one of the basic and most
important processes of memory. But the act of recollection can be
a tough chore.
One way to discover an individuals level of acuity in conjuring up

Memory Study does just that.
A critical part of the Study is the Groton Maze Recall task, or GMR.
It asks participants to reproduce the maze-like pathway (remember
the tile grid?) they were introduced to during the earlier, learning
task phase. Subjects taking Prevagen performed excellentlywith
a dramatic 29% reduction in errors over the 90-day study. Compare
this to the placebo group which only showed a 4.4% improvement,
and you have a compelling reason to take Prevagen!

A Life of Quality

Have you ever heard something, and it made an impression on you?


That is verbal learning. The act of remembering is primal. And
the ability to summon information previously heard or seen is at
our core. Being able to recall (and repeat) something that occurred
earlier gives our lives greater meaning and purpose.
An example might be experiencing something someone saidwords
from a colleague or an actor in a film. Those wordsanything from
something profound and memorable to a joke, made an impression.

When the International Shopping Recall List test was presented
in the Madison Memory Study, the list was read aloud to subjects
three times in succession. Then the other tests were taken. When
completed, testers asked each subject to recall verbally what was on
the shopping list. The results were significant. Over the 90-day run
of the study, subjects within a normal cognitive range and those with
mild to moderate impairment fared well. Both groups had taken
Prevagen during the three-month study period.
While little in this world is perfect, research, executed properly,
uncovers ways that can improve the quality of our lives as we age.
And being able to remember and repeat something verbally to others
is an example of that quality.

In the Madison Memory Study, the Prevagen arm significantly

In an earlier trial, The Prevagen Quality of Life Study tracked


generally healthy participants over a 90-day period measuring
changes in overall cognition, quality of sleep, energy, mood, and
general health. Changes in performance were measured, through

Epilogue
Researchers Discover Key to Brain Health

ne evening I came across a very interesting article in a scientific publication.


It stopped me in my tracks. It said that an Australian swimmer who had
been stung by a jellyfish suddenly developed neurological symptoms. This
really piqued my interest.

How could the jellyfish have this poison inside of it, without
suffering from any effects? What was it in the jellyfish, could this
jellyfish protein help to protect support our brains in the same way
as the jellyfish? This sounded a lot like the snake and its venomwhy
doesnt the snake get killed by its own bite?
The brain and how it works became a topic of interest to me at a
young age because I was forced to face what happens when the brain
doesnt work. A family history of memory problems has led me to a
lifelong study of the brain and how we can help to keep it working.
In college, as a student at the University of Wisconsin at Milwaukee,
I focused my studies on neurochemistry and neuropsychology. What
I found was really exciting. Extensive research on a rare species
of jellyfish had been done and the amazing properties a protein
produced in its body gave it its ability to glow in the dark.
Recently, this experience inspired me to write a book titled, Gift from
the Sea How a Protein from Jellyfish Fights the Aging Process. I hope that
people will find the connection between our brain health and these
unique sea creatures that glow in the dark to be as fascinating as I do.
Together with Mike Beaman we founded Quincy Bioscience in 2004
to further develop the jellyfish technology to be used for supporting
the brain.*

The Effects of the Calcium Binding Protein Apoaequorin


on Memory and Cognitive Functioning in Older Adults
SUMMARY
Background

We report here on a double-blind, placebo-controlled study designed to


assess the effect of an apoaequorin supplement, compared to placebo, on
specific areas of cognitive functioning using quantitative, computerized
assessments. In a recent double-blind, placebo-controlled trial, older adults
taking an apoaequorin supplement reported improvements in cognitive
functioning. Apoaequorin, a calcium-binding protein, is a unique main
ingredient originally isolated from the jellyfish, Aequorea victoria.

Methods

A total of 218 adults, aged 40 to 91 years, who had self-reported mild


memory concerns were randomly assigned to receive a 90 day supply of
either apoaequorin 10 mg daily or a matched placebo. Participants were
tested at predetermined time points using computer-based assessments
from CogState Ltd (www.cogstate.com). Changes on specific assessments
of cognitive function were measured at various time points during the
study.
Results

The apoaequorin arm showed a significant improvement in scores of


executive functioning on the Groton Maze Learning task over the 90day study period. In measures of learning, study participants given
apoaequorin demonstrated significantly improved recall performance at
Day 90 compared to Baseline.
In an assessment of short-term memory and learning, the apoaequorin
arm saw significant improvement at Day 90 compared to baseline in the
One Card Learning task. The apoaequorin group also improved their
ability to recall shopping list items from a previously presented list in
the International Shopping Recall List test. Apoaequorin was very welltolerated in this study.
Conclusion

These results indicated a strong relationship between apoaequorin and


improvements on several quantitative measures of cognitive function.
Apoaequorin has been shown to be a well-tolerated and effective
supplement for use by adults as they age. These results suggest an important
role for apoaequorin in improving cognitive functioning problems that are
associated with aging.
Methods

All participants completed the AD8 Screening Interview (AD8) prior to


their baseline cognitive testing. Participants were additionally required to
complete five (5) cognitive testing sessions. The CogState Research Battery

(CogState Ltd.) was utilized and included measures of executive function,


speed of visual processing, psychomotor function, attention, verbal learning,
delayed recall, and simple and complex working memory. Cognitive testing
sessions lasted between 35 to 60 minutes depending on the participant,
and were administered by a trained researcher at predetermined intervals
throughout the three (3) month study.
Methods

The primary efficacy variable measured the change from the Baseline/
Day 0 to Day 90 as recorded by the CogState software. The measurements
included speed of performance, total number of correct moves per second,
total errors, and the accuracy of performance. The secondary efficacy
variables were self-reported improvements defined by positive changes
from Baseline/Day 0 to Day 90 on the qualitative survey instruments.
Data Analysis: Analysis was conducted using the IBM Statistical Package
for the Social Sciences (SPSS) version 19 (IBM, Inc.). Data from all 218
participants were included in the analysis. The qualitative survey data
were analyzed to find descriptive statistics, the Mann-Whitney U Test and
the Wilcoxon Signed Rank Test. Group means and standard deviations
were found for each cognitive assessment. Each cognitive assessment was
analyzed using paired and independent t-tests, and the Mixed-Model
Repeated Measures Analysis of Covariance (ANCOVA). The Baseline/
Day 0 test scores served as the covariate for the Mixed-Model Repeated
Measure ANCOVA. Participant data was segregated for analysis based
upon self-reported level of impairment as measured by the AD8 score prior
to Baseline/Day 0.
Participants: The Madison Memory Study sample was comprised of
218 participants (148 females and 70 males) aged 40 to 91 (= 62.48) years.
Eligible participants were randomized, by a 2:3 ratio, into the Placebo arm
(n=92) or the Apoaequorin arm (n=126) respectively.

Study Design: The Madison Memory Study was a double-blind,


placebo-controlled study. Participants were randomized into the Control
arm or the Experimental arm with a random number generator using
a 2:3 ratio. In the Control, or Placebo arm, participants received a
supply of capsules containing only white rice flour. Participants in the
Experimental, or Apoaequorin arm, received a supply of capsules
containing 10 mg of Apoaequorin in addition to the white rice flour.
Placebo capsules were size and color matched to the Apoaequorin
capsules. Participants were required to take one (1) capsule daily for the
entire duration of the three (3) month study.

Primary Objective: To compare and assess the effectiveness

Whereas, the GMR task reported the total number of errors made
during the participants recall of the same maze seen previously in

Additionally, participants in the Apoaequorin arm within the
normal cognitive range (0 to 1) on the AD8 showed significant
change over time on the GML from Baseline/Day 0 to Day 90, (=
12.81, SD= 18.47), t36= 4.29, p< .000, Cohens d= 1.00. There
was a decrease in total errors by 22.88% over the course of the
study.

A significant difference was seen over time between the


Apoaequorin arm and the Placebo arm in the total number of
errors made on the GMR for participants who scored within
the normal cognitive range (0 to 1) on the AD8, F1, 54 = 7.19,
p< .01. Additionally there was a significant effect seen within
the Apoaequorin arm when comparing the Baseline/ Day 0
and Day 90 results, (= 8.92, SD= 20.33), t36= 3.67, p< .001,
Cohens d= 0.8.
The total errors in the Apoaequorin arm for participants who
scored within the normal cognitive range (0 to 1) on the AD8,
between Baseline/ Day 0 and Day 90, decreased by 29.12% on
the GMR compared to only 4.35% in the Placebo arm (Figure
2).
One Card Learning Results (Figure 3)
One Card Learning:
The One Card Learning (OCL) test measured the proportion of correct
responses during the entire duration of the test. Participants were required
to remember each card that was shown in addition to responding correctly
as to whether a card was new or had been seen previously.

The Apoaequorin arm saw a significant change from Baseline/Day


0 to Day 90, (= -0.03, SD= 0.14), t126 = -2.08, p< .05, with 61.47%
of participants showing an improvement from the Baseline/Day 0 to
Day 90 as measured by the accuracy of performance (Figure 3). The
accuracy of performance is the arcsine transformation of the square
root of the proportion of correct responses.
International Shopping Recall List Results (Figure 4)
The International Shopping Recall List:
The International Shopping Recall List (ISRL) measures the number
of correct shopping list items the participant can recall from a
previously presented list of items. Verbal learning and delayed recall
are cognitive functions specifically measured by the ISRL.

A significant effect was also seen over time on the ISRL for
participants within the mild to moderate cognitive range (2 to
5) on the AD8 in the Apoaequorin arm, (= -2.76, SD= 5.41),
t67= 2.40, p< .05 (Figure 4).
Summary
Overall, participants in the Apoaequorin arm and within the
normal to mild cognitive range on the AD8 saw a significant
positive change over the three (3) month study period in:
Verbal Learning (ISRL)
Memory (OCL)
Delayed recall (GMR and ISRL)
Executive Function (GML)
Conclusion
This study establishes a relationship between Apoaequorin and improvements
on quantitative measures of cognitive function. These results show statistical
significance between Prevagen and improvements on several quantitative
measures of cognitive function. Prevagen has been shown to be a well-tolerated
and effective dietary supplement for use by adults as they age. These results
suggest an important role for Prevagen in improving cognitive functioning
associated with aging.
