Project is a word that terrifies most of the college students. This is because of the experience they had in
their schools. I remember when I searched a lot for project ideas as my project guide and other
professors werent that supportive. So here are a few innovative Micro biology project ideas to help you
out:

Immunology and Antibodies


You could compare the antibiotic activity of a certain product both in-vitro and in-vivo.
Red Tide
You can consider the algae species and analyze the toxins released by them and the conditions under
which they release the toxins. In particular, shellfish-related poisoning in humans caused by red tide
toxins is common, so a project can explore the relationship between the algae and human food sources.
Bacterial Sensitivity
Bacteria are sensitive to ultraviolet rays, which can cause variations of particular bacteria in different
ecological situations. Consider a project that measures the effects of these rays on different bacteria.
Variables known to affect the outcome of this "bacterial sensitivity" include the wavelength used, the age
of the bacteria and the temperature the experiment is carried out in. Apply your laboratory findings to real
world situations in order to posit the relationship between bacteria variations and ultraviolet rays.
Water Microbiology
You can measure the microbial growth in municipal water supplies, well water or naturally occurring
aquatic ecosystems. This will also come under waste water treatment.
Antibiotic Resistance:
You can also measure the resistance of various microbes and make a report about the superbugs.

Phage inside human cells
Bacteria that invade human cells buy time to acquire antibiotic resistance genes. We showed that
invasive bacteria exchange genes by conjugation inside human cells, making the cytoplasm of human
cells a potentially highly productive ecosystem for gene transfer. Now we will determine how phage enters
the cytoplasm of human cells and assess the use of phage as therapeutics to control antibiotic-resistant
pathogens. We will use sophisticated genetic techniques along with confocal and epifluorescence
microscopy to:
1. define the place within the cell that pathogenic bacteria and phage meet,

2. test virus killing of antibiotic resistant bacteria inside human cells,


3. determine what happens to phage and bacterial nucleic acids released into human cells.

Background/skills required: Required: enough chemistry to make buffers and an understanding of
microbial genetics. Desirable: understanding of bacteriophage and tissue culture techniques

Microalgae

Taxonomic studies on cyanobacteria


The diversity of cyanobacteria in New Zealand is poorly known in almost all habitats. Only freshwater
phytoplankton communities have been examined in any detail. Thorough examination awaits for those in
soils, on rock surfaces, in benthic marine and freshwater habitats and associated with fungi in lichens.
The aim is to provide excellent descriptions of taxa. This would provide essential base-line knowledge to
support subsequent studies in a wide range of disciplines, e.g. ecology, use of cyanobacteria as
environmental indicators, investigation of bioactive products. Descriptions would be based on field and
cultured specimens. Light microscopy, transmission and scanning electron microscopy would be used.
Molecular genetic approaches could be applied if a supervisor for those could be found. The study could
experimentally investigate the stability of phenotypic characters in selected taxa. Unusual, barely studied
and novel taxa would almost inevitably be encountered.

Soil Biology
Diversity and distribution of soil algae
There have been very few investigations of soil algae in New Zealand although they are an important
component of microbial communities in the upper few centimetres of most soils. How diverse are they?
How do communities differ between naturally-occurring and agricultural and other human-impacted soils?
Are New Zealand soil algae very similar to those described from other regions or do some soil types
support unique communities? Do high alpine soils support communities similar to those found in
Antarctica? What controls differences in community structure between different soil types? How
abundant are soil algae? How responsive are they to changes in their microenvironment, e.g. water
availability, temperature, nutrients? A range of taxonomic, ecological and experimental studies could be
designed to suit interests.

Soil Microbiology and Biochemistry


Foams are biological links between terrestrial aquatic systems
Foams occur in tropical and polar regions yet their biology and biogeochemistry is little known. Foams
contain very large concentrations of nutrients and numbers of microbes particularly bacteria. Foams may
function as inocula and are involved in colonisation of soils, at least, in Antarctica. Little is known about
the microbial species present in foams, their survivability and role, if any, in transfers between different
ecosystems.

ability to live in remote, isolated places.

Virology

Australasian Mastreviruses: Diversity, Evolution and recombination


analysis
Mastreviruses are single stranded DNA viruses that infect grasses. Other than four isolates from
Australasia we have very limited information about these viruses and their infectivity in the region. In
African species of mastreviruses we have observed a high level of recombination between strains and
within strains. We aim set up an initial sample set of at least 50 infected samples from which we will
amplify (using rolling circle amplification), clone, sequence and analyse for recombination in the full length
genomes. This project will link in with a larger project run in Africa and the Indian Ocean Islands by
myself, Dr Darren Martin (University of Cape Town, South Africa), Dr Dionne Shepherd (University of
Cape Town, South Africa) and Dr Jean-Michel Lett (CIRAD, La Reunion) which is the largest non-human
pathogen diversity project in the world.

The hunt for novel DNA viruses: Diversity and evolution through recombination
Rolling circle amplification using Phi29 DNA polymerase has revolutionised the non-specific amplification
of circular DNA viruses. Techniques established by my research group at the University of Cape Town,
South Africa, will be used to sample for novel DNA viruses in various ecosystems. Initially the project will
focus on the following:
1. Avian DNA viruses, mainly circoviruses and polymoviruses, in archived samples of mallards and
ducks undergoing an evolution bottle neck.
2. Corticoviruses and microviruses infecting bacteria will involve in dept studies on evolution through
recombination, measuring recombination hotspots and evolution rates
3. Plant viruses such as caulimoviruses and begomoviruses.
