ABSTRACT: The productivity of swine industry is highly dependent on the production and
transport of boar semen. For this reason, developing an extender that preserves semen for
long time is a major goal in swine reproductive technologies. The present study was
performed to evaluate the potential of extra-long term extender Duragen to keep the
fertilization capability of semen after 1, 12 and 15 days of preservation.
The sperm rich fraction of fertile boar ejaculate was collected, evaluated and pooled. Seminal
doses of 3 billions spermatozoa in 90 ml were prepared and stored at 17C before being
used for 1, 12 and 15 days for Duragen and 1 day for BTS extended semen. 84
multiparous sows were randomly divided in four groups according to semen processing: B1
(control group); D1; D12; D15. Cervical insemination was performed at 0 and 24 hours after
the observance of external estrus signs. Pregnancy rate (PR) was evaluated 21 days after
insemination. The sows were slaughtered at 35 days of pregnancy, their genital tracts were
removed and total embryos (TE) were counted.
Pregnancy rate values were clearly compatible with farm production in BTS1; D1 and D12
groups (100%; 100% 75.6%). Even sows inseminated with 15 days preserved doses (D15)
showed PR values over 50 %. No significant difference was found in TE between BTS1, D1
and D12 (13.3 1.17; 15.4 0.88; 14.4 1.14) but semen storage for 15 days clearly
decreases this variable (p<0.005) (9.1 0.85).
In summary, Duragen extra-long semen extender maintained a high degree of sperm
fertility for more than 12 days. On the other hand, data suggested that using Duragen as
short term extender may increase the farrowing rate in farm conditions.
