The Effect of Temperature on Enzyme

INTRODUCTION:
1. Quoting to Mr. Bill Pham, Some chemical reactions that make life possible are too slow to
make them possible for living tissue. These chemical reactions are made possible by catalysts. A
catalyst is a substance that speeds up the rate of a chemical reaction. Enzymes are proteins that
act as biological catalysts. Enzymes speed up chemical reactions that take place in cells.
Enzymes are very specific, generally catalyzing only one chemical reaction.
2. Quoting to Mr. Bill Pham, The Enzyme Substrate Complex: enzymes provide a site where
reactants can be brought together to react. The reactants of enzyme-catalyzed reactions are
known as substrates. Because they are catalysts for reactions, enzymes can be affected by any
variable that influences a chemical reaction. Cells can regulate the activities of enzymes in a
variety of ways. Most cells contain proteins that help to turn key enzymes on or off at
critical stages in the life of the cell. Enzymes play essential roles in regulating chemical
pathways, making materials that cells need, releasing energy, and transferring information.
Enzymes help the reaction fatser.
3. Quoting to Mr. Bill Pham, Factors that influence enzyme activity are: pH, temperature, and
other proteins (we call them regulatory proteins), and some factors such as enzyme
concentration, presence of any inhibitors or activators, and substrate concentration.
4. The purpose of this lab experiment helps us to understand about enzymes, and some factors
that is effect the enzyme productivity. Figure out how the pH work in room temperature, hot
temperature, cold temperature, and how it effect on enzymes. We can study about the effect of
temperature on enzymes activity.
5. The enzyme produced bile in liver. Quoting to the Reference, Hydrogen peroxide also has
antiseptic and antibacterial properties that make it useful for a variety of medical and cosmetic
purposes. However, the chemical has to be severely diluted for uses, as solutions containing
more than 8 percent hydrogen peroxide are corrosive to the skin. Most commercial grades of the
chemical contain somewhere between 35 and 90 percent hydrogen peroxide in a solution of
water and trace amounts of stabilizers. We use hydrogen peroxide to clean a wound from not

being infected. Especially, we cannot drink the hydrogen peroxide because it is a chemical, it is
harmful, and it may kill you.
6. If we test catalase in a cold condition, the rate at which catalase breaks down H2O2 will be
slower. If we test catalase in a hot condition, the rate at which catalase breaks down H2O2 will be
faster.

MATERIAL:
-Liver

PROCEDURE:
For each experiment, you need to do for 3 times
1. Put on gloves.
2. Grind liver.
3. Put the grind liver into the petri dish.
4. Take the standard scissor to cut the filter paper so that it can fit in the beaker.
5. Now, test it with room temperature, hot and cold temperature.
Hot Temperature:
-Take the bath, and pure the hot water into it.
-Put the petri dish (make sure the petri dish that you already put the grind liver into it) into the
bath.
-Take the thermometer to measure the temperature of the liver.
-Put 25ml of hydrogen peroxide H2O2 into the beaker.
-Pick up the filter paper, and put the filter paper on top of the liver petri dish to attaching the
enzyme (just only the little amount). Then, take it out.
-Put that filter paper into the beaker (already have hydrogen peroxide H2O2) push that filter
paper until it reach the bottom of the beaker.
-Take the time measurement out to measure when it starts bubbling until it stops bubbling.
-Write down the measurement that you measure into your data table.
Cold Temperature:
-Take the bath, and pure the water into it.
-Put ice into the bath.

bath.

-Put 25ml of hydrogen peroxide H2O2 into the beaker.

3. To my observation on the graph data, for the slowest one is cold temperature; it bubbled with a
small amount of bubble, and slowly. Room temperature is faster than cold temperature; it
bubbled faster, and bubble with a larger amount than the cold temperature. For the fastest one is
hot temperature; it will take the shortest time to bubble, and bubbled with a large amount of
bubble.

DISCUSSION:
1. The trend of my graph go from the highest to the lowest; it starts from the left to the right. It
bases on the time so we can know that which one is the fastest, and which one is the slowest. It is
important for you to graph the data because we can get the visual image, to observe clearly, and
is it growth or develop. Without the graph, it wouldnt be easy for us to describe the trend by just
looking at the data table alone.
2. Yes, it help me in the hypothesis because at first our hypothesis is if we test catalase in a cold
condition, the rate at which catalase breaks down H2O2 will be slower. If we test catalase in a hot
condition, the rate at which catalase breaks down H2O2 will be faster. It helps me realize that my group
hypothesis is right, ad match with the graph.
3.measure the wrong time, too much of the liver in the filter paper, measure the wrong temperature are
those could be some experimental errors that affected your results.
4. We repeat the experiment three times because we want to see is it change or not, and also to see the
exactly result of the experiment.

5. We have to take the average of these trials for each testing condition to know the accurate
number of the result because we do this experiment for three times; so we need to take the
number of the result after three times plus it together and divided by three to have the average.

RESEARCH QUESTIONS:
1. Quoting to the Quizlet, Enzymes fit together like a lock and key. Enzymes are catalysts
because they speed up the rate of chemical reactions. When the substrates go in the enzyme, they
form an enzyme-substrate complex.
2. Quoting to the Quizlet, A change in pH could change the shape of hexokinase. This change
would diminish or possibly eliminate the ability of glucose and ATP to bind to the active site on
the enzyme.

REFLECTIONS:
1. I can learn that grind liver is very stinking. I can learn how to make a data table, and the graph
of data. Without graph data, it wouldnt be easy to describe the trend by just looking at the data
table alone. Lab helps me to study and understand more about biology. I can learn how to do a
lab report. Team work is very important in lab, and work with team you can do everything
quickly in a shortest time.
2. This experimenting was great and I like it a lot. It is fun when you work with your team.
3. Yes, I do. They are very collaborative; each of them knows what they are doing. We help each
other by working together, and divide work for each other to do. There are three people who
measure the temperature, and pick up the filter paper. Two other are in the table, one is clean up,
and put 25ml of hydrogen peroxide H2O2 into the beaker to be ready for the three people; another
one is measure the time, and write down the data (time, temperature, hot, cold, room
temperature). We are all observing the effect by the calling hey, groupgroup.. of our
teammate.
4. Sometime, we are still dont understand each other. So next time, we will try our best by
listening to each other
