Soft capsules are a single-unit solid dosage form, consisting of a liquid or

semisolid fill enveloped by one-piece hermetically sealed elastic outer shell.

One of the major challenges in the
development of soft capsules is that the
system is very dynamic in terms of physical
migration or even chemical interactions.

If formulation or process is not well-done, migration can continue through shelf life:

Water migration does not continue through stability.
Results affected by uniformity of dosage
We can test the whole capsules but variability affects assay, but not CU.

Conclusions

Conclusions
Soft gelatin capsule is a unique dosage
form where dynamic equilibrium affect both
shell and fill composition.
Final equilibrium should be reached after
drying, but depends on formulation and/or
process, it can continue through shelf-life.
Analytical approaches need to take into
account migration phenomena, in order to
avoid either low or high variable results.
According to the approach, strategy for
validation need to be adjusted, in order to
actually verify the capabilities of the
proposed method for the intended purpose.
