carcinogeneihaeenneitherundertoodnorexplainedforalmotacentur.Warurgtartedthefieldofcancermetaolim,whichecamethepredominant
fieldincancerreearchforeveraldecadeafter.However,theentirereearchincancertookadramaticturnuponthearrivalofgeneticwhenin1953Noel
laureateJameWatonandFranciCrickdicoveredthetructureofDNA.the1980andonward,cancerreearchwafullemeddedinthegeneticfield
aroundtheworldduetotheexcitementofgeneticandallofitpoiilitietocuredieae.Inthemeantime,cancermetaolimwaforgottenanduriedatthe
ottomoftheocean.

Unfortunatel,thefightagaintcancerolelthroughgenetichaproaleenoneoftheiggetmitakeinmedicalreearchhitor,afterhalfacenturof
reearchandhundredofillionofdollardeploed,thecureagaintcancerthroughgeneticremaineluiveandfarawafromarealit.JameWaton
himelf,whotartedthefireincancergenetic,inowcriticizingthefieldfornotlivinguptotheexpectation,tatingthatlocatingthegenethatcauecancer

thelactateretoredtoglcogenduringaeroicrecover.In1923anotherNoelLaureate,ArchialdVivian(AV)HillandhicolleagueWalterMorleLupton
decriedthetermO2Detinwhichthelinkedlactateproductionduringexercietooxgenlimitedlactateproduction.uequentl,Warurgatutel
decriedthatlactateproductionincancercellwaduetoaninjurtomitochondria,whichwemacalltodamitochondrialdfunction.However,neitherthe
knowledgeofgeneticnortheufficienttechnologietotudlactatemetaolimwereavailaletoWarurgandhicontemporarieduringhitime.Further,
ecaueofthetatureofthefirtinvetigatorinlactate,theconceptoflactateproductionaareultofoxgenlackandaawateproductwaimmortalizedin
textookofphiologandiochemitrforaoutacentur.
However,inthemid1980GeorgerookfromtheUniveritofCaliforniaaterkele,decidedtounurlactatefromtheottomoftheoceanandemarkedon
afacinatingadventuretoundertandlactateanditroleinkeletalmucleduringexercie.Overtheear,Georgerookhaeenaletoprovethatlactatei
notawateproductutahighlactivemolecule,oneofthemotimportantgluconeogenicprecuror,amajorutrateforalmotevercellintheod
includingtherainortheheart,amajorregulatorofintermediarmetaolim,epeciallfattacidmetaolim,ahighlactiveignalingmoleculewithhormone
likepropertie,andproalevenatrancriptionfactor.

WhenIfirtreadaouttheWarurgffectincancerovertwoearago,Iwaurpriedtoeethatitwanotaoutgeneticutaoutmetaolim.
Neverthele,IwamoreurpriedwhenIawthatitwathemainujectofmdoctoratetheialthoughinkeletalmucle,wecallitctoolicglcolior
implglcoli(underaeroicoranaeroiccondition).Theplaerofglcoliothincancerandkeletalmuclearetheameexceptthatincancercell
thearecompleteldregulatedproalaprimarilgeneticdregulation.Inkeletalmucle,however,glcoliplaerareperfectlregulatedwherein
