When Food Meets Man: the Contribution of Epigenetics to Health

Abstract: Post-translational modifications of chromatin contribute to the


epigenetic control of gene transcription. The response to food intake and
individual nutrients also includes epigenetic events. Bile acids are necessary for
lipid digestion and absorption, and more recently have emerged (have been
identified) as signaling molecules.
Their synthesis is transcriptionally regulated also in relation to the fasted-to-fed
cycle, and interestingly, the underlying mechanisms include chromatin remodeling
at promoters of key genes involved in their metabolism.
Several compounds present in nutrients affect gene transcription through
epigenetic mechanisms and recent studies demonstrate that, beyond the well
known anti-cancer properties, they beneficially affect energy metabolism.
Obese subjects have high expression Leptin secretion
and low Adiponectine from adipose tissue.
Lean subjects and even fasting obese subjects have high Adipokine
expression and secretion by adipose tissue
IMPROVING INSULIN RECEPTORS

In diabetes type 2 the number of receptors to insulin on the


lymphocyte is half that of non-diabetics. Diabetics with obesity
are nearly one third of non-diabetics.

to have less glucose and less insulin!

Fasting for 72 hours also protected cancer patients against the toxic impact of
chemotherapy.
Neuroprotection
IF up-regulation of neurotropic factors like Glial cell-derived neurotropic factor
(GDNF) and brain- derived neurotropic factor (BDNF).
These both promote survival of neurons as well as the making of new connections.

Chaperone proteins like shock protein-70 (HSP-70) and glucose-regulated protein


78 (GRP-78) are also up-regulated, these can protect the cells from damage and death
from environmental conditions.

Cytoprotective activity with regard to seizures and epilepsy


IF has been shown to be more efficient that CR in the previously mentioned ketogenic
metabolic pathway.

This ketone body exhibits neuroprotectivity in models of Alzheimers and


Parkinsons disease (Kashiwaya et al., 2000). IF fed mice can develop a twofold
concentration of the -hydroxybutyrate ketone body compared to mice fed ad libidum
(Anson et al., 2003).
Intermittent fasting (IF) induces the
expression of protein chaperones (e.g.,
heat-shock protein 70 and glucose-
regulated protein 78) and stimulates
autophagy in many cell types
(Duan and Mattson, 1999; Yu and Mattson, 1999; Alirezaei et al., 2010;
Arumugam et al., 2010; Lee and Notterpek, 2013).
In addition to being used as an energy source, ketone bodies can
modulate signaling pathways and gene expression by, for
example, inhibiting certain histone deacetylases
(Newman and Verdin, 2014).

Fasting can enhance the ability of cells to damaged proteins and


organelles, a process called autophagy (Jamart et al., 2013).

The latter actions of intermittent fasting (IF) enhance the


ability of cells to eliminate damaged proteins and organelles
such as mitochondria.
Animals that eat only every other day live up to
30% longer than those that eat every day
(Goodrick et al., 1983), and exhibit
resistance to neurodegenerative disease
(Duan et al., 2003; Halagappa et al., 2007;
Arumugam et al., 2010).
During the fasting period, fatty acids are released from adipose
cells and enter the liver where they are converted to ketone
bodies (-hydroxybutyrate and acetoacetate) which provide a
robust energy source by for muscle cells and neurons
(Johnson et al., 2007; Maalouf et al., 2009).

Ketone bodies have beneficial effects on cells that can counteract


disease processes.

For example, ketones protect neurons against dysfunction and


degeneration in experimental models of epilepsy
(McNally and Hartman, 2012) and Alzheimers disease
(Kashiwaya et al., 2013).
Intermittent fasting (IF) can also increase
neurotrophic factor signaling in brain cells,
which may contribute to their abilities to
enhance hippocampal neurogenesis (Lee et al.,
2002) and protect neurons against oxidative
and metabolic stress in animal models of
Parkinsons disease (Duan and Mattson, 1999),
Huntingtons disease (Duan et al., 2003), Alzheimers
disease (Halagappa et al., 2007) and stroke (Yu and Mattson,
1999; Arumugam et al., 2010).

The role of nitric oxide (NO)
for effective treatment of microvascular complication

Normoglyceamia is most important condition!
Hyperglycaemia impairs the activity of macrophages to
produce nitric oxide (NO), absence of NO leads to vasoconstriction.

Vasoconstriction cannot appear if is hyperglycaemia.

IHT cannot be enough effective if is hyperglycaemia.

Impairs the activity of NO resulting in endothelial dysfunction: causes


vasoconstriction, smooth muscle proliferation, platelet activation/aggregation,
and leukocyte adherence to the endothelium

Cold water adaptation increase in brown fat cells, which protect the body from
aging. Brown-fat cells have a large concentration of mitochondria to generate heat
without physical contraction or muscular movements.

One of the important pathogenic factors in cold therapy is the release of nitric oxide
(NO), leading to a rapid increase in skin blood flow. Hyperemia is largely (70%)
governed by the endothelial release of nitric oxide (NO).
