Abstract
Introductions: Mortality caused by hypercholesterolemia is increasing, especially in the
productive age. Hypercholesterolemia could cause various diseases such as coronary
heart disease, stroke, and atherosclerosis. Cholesterol could be formed by LDL
oxidation. It could be prevented by various compunds which have antioxidant activity.
Peanut skin (Arachis hypogaea L.) extract contains various antioxidant compounds;
epicatechin, p-coumaric acid, fleuric acid, chlorogenic acid, quercetin, and p-
hydroxybenzoic acid. This study aimed to test the anti-cholesterol bioactivity in peanut
skin extract and its effect on the level of LDL, HDL, triglycerides, and total cholesterol in
the blood, as well as to determine the dose of peanut skin extract that could provide
anti-cholesterol effect.
Methodology: Ethanolic extract obtained by macerating 1 kg peanut skin using 96%
ethanol during 5x24 hours. Then in vivo test of anti-cholestrol activity trials were done
with male rats aged 8-10 weeks. The rats were divided into 6 groups (6 rats each). First
group was pellets fed, without treatment extract. The second group was fed a high-fat
diet (HFD) without extract. The third until fifth group were given HFD and extract at a
dose 100 mg/200 g BW, 400 mg/200 g BW, and 1000 mg/200 g BW. The sixth group
was given HFD and anti-cholesterol drug simvastatin (0,18 mg/200 gr). After 30 days
treatments, consentrations of HDL, LDL, triglycerides, and total cholesterol in blood
were tested. The data obtained were processed using ANOVA Oneway and Kruskal
Wallis (confidence level 95%).
Results & Discussion: The rendemen that we got from maceration 1 kg of peanut skin
is 3.381%. The in vivo test showed peanut skin extract reduced triglycerides and total
cholesterol levels while it didnt have any significant influence on LDL and HDL level.
The group which has the most significant influence to triglycerides and total cholesterol
is group with treatment dose 1000 mg/200 g BW.
Conclusion: Ethanol extract of peanut skin (Arachis hypogaea L.) reduced triglycerides
and total cholesterol levels at 1000 mg/ 200 g BW dose of extract.
