ABSTRACT
Background: Umbilical cord-mesenchymal stem cells (UC-MSCs) accelerating
wound closure by increasing VEGF and PDGF level leading to re-epithelialization,
cell infiltration, and angiogenesis. It has been found that MSC-conditioned medium
(MSC-CM) can enhance migration of fibroblasts in scratch assays. However, the
effect of MSC-CM with Serum Injured in accelerating wound healing remains
unclear. This study aims to evaluate effect of several doses topical MSC-CM in
accelerating wound healing.
Method: Eighteen Wistar rats were randomly assigned into vehicle group (Veh),
treatment group (T1, T2) and were received serum-free gel (Veh), MSC-CM gel 1:5
(T1), MSC-CM gel 1:10 (T2) respectively, twice daily for 9 days. PDGF and VEGF
level measured using ELISA on day 3 and 6.

Result: This study showed a significant increase in VEGF and PDGF level (p<0.05)
on day 3 and 6, along with a significant increase (p<0.05) in fibroblast density. 

Conclusion:   In   conclusion,   topical   gel   with   high   concentration   of   MSC­CM   is


effective to accelerate wound closure by enhancing PDGF and VEGF level in full­
thickness skin defect rats compared to control group.
