ABSTRACT
Nurhayati Pratama Andreyansah. 2010. Antagonistic Potential of
Endophytic Bacteria from Potato Roots and Tubers on Potato Cyst
Nematode (Globodera rostochiensis Wolleweber) In Vitro. Supervised by Yadi
Supriyadi and Noor Istifadah.
Globodera rostochiensis is an important pathogen in potato crops in
several regions in Indonesia. One of the control measures need to be developed is
biological control. Endophytic bacteria are potential as biological control agents
of plant parasitic nematodes. The purpose of this study was to obtain non
pathogenic isolates of endophytic bacteria that their culture filtrate are
antagonistic to G. rostochiensis in vitro.
The experiments were conducted at Laboratory of Nematology and
Laboratory of Phytopathology in the Department of Plant Pests and Diseases,
Faculty of Agriculture, Universitas Padjadjaran from May to September 2010.
Potato samples were obtained from several areas in West Java which were
Bandung (Pangalengan and Ciwidey), West Bandung (Lembang) and Garut
(Cisurupan, Pasirwangi, Cikajang). This study was conducted in several stages,
which were isolation of endophytic bacteria from the potato roots and tubers,
testing the influences of the isolates on potato plants and the non-pathogenic
isolates were further tested for their antagonistic effect on G. rostochiensis in
vitro. The experiment was arranged in Completely Randomized Design (CRD)
with endophytic bacterial isolates and control as treatments. Each treatment was
repeated 3 times.
Among 88 isolates of endophytic bacteria obtained from potato roots and
tubers, there were eight non pathogenic isolates that their filtrate caused
82.52% - 97.92% Juvenil 2 G. rostochiensis damaged. All the isolates tested were
not effective in causing damage to the egg of G. rostochiensis.
