Evaluation of Anti-angiogenic Activity by Chick Chorioallantoic Membrane Assay of Commercially Available Food Supplements

ABSTRACT

Angiogenesis is the process of developing new blood vessels from existing blood vessels. The inhibition of this process is called anti-angiogenesis, which was the main focus of the study. Studies showed that garlic oil, green tea and Shiitake mushroom extract inhibit angiogenesis, which was the basis of choosing dietary supplements containing these extracts as the samples. The study aimed to determine if the consumption of these dietary supplements causes anti-angiogenesis and which of these samples present the most visible anti-angiogenic effect. It also aimed to determine if there is a significant difference between the samples and the positive& negative controls which are cyclophosphamide (Endoxan) 200 mg and 0.9% Sodium chloride respectively. Fertilized Philippine duck eggs (Anas luzonica) were used as test specimens to confine the samples and were incubated at 37C at constant humidity for eight days. On the eighth day of incubation, a small hole was made on the position where the air sac is located. Through the 1cmx1cm window, the test sample was confined on the CAM using Whattman filter disc then covered with the shell previously removed then the filter paper and Parafilm sheet thereafter. The eggs were incubated for three more days. On the eleventh day of incubation, the eggs containing the samples were photographed and compared to the control eggs. Inhibition of angiogenesis was evaluated thereafter. For the statistical analysis, ANOVA was used followed by multiple range tests and a certified statistician was held responsible for these matters. According to the statistical analysis the variances across groups vary. From the samples, only shiitake has a significant difference with the negative control and no significant difference with the positive control. Garlic oil and green tea did not have any significant difference with the results of the negative control, while having significant difference with the positive control. To conclude, not all of the three dietary supplements caused an antiangiogenic effect and shiitake presented the highest antiangiogenic activity.
