Our History Orochem Technologies Inc., established in 1996, is a biotech company that serves the bioanalysis, genomics, drug discovery, and the proteomics markets. Moving beyond its established reputation as the pre-eminent supplier of sample preparation tools, the acquisition of Column Engineering Inc in 2006 provided an opportunity for us to service the varied HPLC market. Since then Orochem Technologies has greatly broadened its expertise and manufacturing capabilities in HPLC products and services. Mission Statement At Orochem Technologies, the core expertise is in the development and manufacture of customized individual and high throughput sample preparation devices, unique chemistries for columns, analytical, preparatory, and chiral chromatography. We are driven to understand and offer tools for parallel processing, scale-up and process chromatography. Our mission is to offer a one-shop to integrate indigenously developed chemistries for separations into custom plastic molded or machined devices. Knowledge of chemical biochemical processes assay sample preparation system allows us to integrate and support sample prep and chromatography systems into automated robotic workstations.

They function as a filter and adsorber of materials that can adversely affect the chemistry of the separation or the lifetime of the column. Their use is highly recommended when injecting anything other than pure filtered standards that elute in a reasonable number (<20) of column volumes. The best way to correctly choose the appropriate guard cartridge is to first consider its objective in the system. There are a multitude of designs and sizes available, and proper choice will provide maximum benefit with minimal effects to the chromatography. Keep in mind that guard cartridges are small columns, and as such they will have backpressure and will contribute to the separation resolution. Because of the effect, even though it is minor, that guard cartridges have on the separation, it is always advised that the stationary phase inside the guard cartridge be the same as that in the analytical column to avoid unexpected results. Typically it is best to use guard cartridges with internal diameters comparable to that of the analytical column. This insures that the linear capacity of the cartridge is adequate for the expected column injection sizes and that the contribution to backpressure is minimal. Frequently chromatographers will choose guard cartridges packed with media of a larger particle size than that in the HPLC column. This also helps to reduce the backpressure contribution from the guard cartridge. Since the length of the guard cartridge packed bed is typically less than 1/10th of the HPLC column, its contribution to the overall efficiency and symmetry is negligible unless the difference in particle sizes between that used in the column and that used in the guard cartridge is greater than an order of magnitude. Since the guard cartridge is like a small HPLC column, proper care must be taken in plumbing it into the system to avoid an increase in extra dead volume. Use minimal tubing lengths from injector to guard cartridge and from guard cartridge to column. The most common question regarding guard cartridges is when they should be changed. If the backpressure increases more than 10% beyond that observed with a fresh guard cartridge, change the cartridge. If a noticeable deterioration of the chromatography occurs after several injections, change the guard cartridge. In summary, guard cartridges are a valuable tool, and with proper use they can help to extend HPLC column life significantly without any adverse effects.

Testing the column with the test mix and under release conditions as specified on the release QC chromatogram will reveal whether the problem is due to a poor column or due to poor separation chemistry. Clearly, if the chromatogram obtained from the test mix closely corresponds with the release QC chromatogram, the column is not the source of the problem, and the separation chemistry is the most likely source of the problem. If the chromatogram does not corresponds closely to the release QAC chromatogram, then the column is the source of the problem. Therefore, it is advised that when separation problems arise, simply running the appropriate test mix under prescribed conditions can point to the source of the problem.
