Joni has been in the industry for 23 years, first with Gilson and now Agilent. Joni has maintained a focus on sample preparation not only techniques but automation Being very customer focused and working directly with customers in assisting with their applications has given her great insights into providing solutions to customer application problems. Joni is a very hands-on scientist always trying to broaden her understanding of techniques and applications, working in the lab on a regular basis which allows her to offer customers a real life experience and understanding Joni delivers presentations worldwide to customers and at conferences and regularly offers hands on workshops. Dr Linda L Lloyd: Product Manager BioLC Columns

Linda has been working in the field of Liquid Chromatography since joining Polymer Laboratories Ltd back in 1979 where she started in production followed by positions in research and development, applications and technical support before becoming a technical product manager with global responsibility for the HPLC columns and media. Linda has published over 50 papers in the areas of material development for HPLC and HPLC applications and presented on behalf of her team at many conferences around the world. Lindas current responsibility at Agilent is as Product Manager for the BioHPLC columns. In this role she works with colleagues from across the Agilent organization to integrate the columns into work flow solutions for the characterization of biomolecules and to define the strategy for advancement of biocolumns technology to provide enhanced solutions to address the current work flow demands.

Bradley Skelchy

Bradley graduated with a BSc (Hons.) Biochemistry from University Putra Malaysia. He worked as a Senior Chemist in both QC and R&D in a Generic Pharmaceutical for 7 years and now has spent more than 4 years as a Senior Applications Chemist for Agilents primary consumables distributor, IT Tech Research in charge of troubleshooting, lectures and consultation in application and support for Agilent consumables" Ian Sik Ian Sik has joined Agilent Technologies in 2007 as an applications chemist to support Agilents growing Chromatography and MS customers base in Malaysia. He is currently the Product Specialist under the Chemistries and Supplies division whose task is to manage the growing business of the aftermarket sales in Malaysia. Prior to joining Agilent, he worked as a research officer in the Institute of Medical Research for several years specializing in natural products chemistries research. He is a graduate in the field of Industrial Chemistry.

Optimization of a SPE Approach Through the Use of Method Development Tools Dr Joan Stevens It is important to understand and use method development tools when establishing a SPE method. Basic tools that are available and to be discussed include the use of an organic gradient to determined post sample load optimization. Understanding compound chemical properties; how to exploit those parameters during method development. Explanation of matrix and matrix effects and addressing these issues in regards to compound extraction Column Choices and method optimization for HPLC characterization of biologics Dr Linda L Lloyd Analysis of complex biomolecules requires multiple HPLC techniques, size exclusion, ion exchange and reversed phase. In this presentation we will discuss these three separation mechanisms and how they may be optimized and controlled for biomolecule characterization. The choice of the optimum column geometry and media will be reviewed and correlated with the physicochemical characteristics of the biologic. We will conclude by presenting a number of application examples, with analysis conditions, that can be used as the initial data point for method development. The role of HPLC in the biopharmaceutical development lifecycle Dr Linda L Lloyd As a biopharmaceutical candidate moves through the development process the requirements and demands placed on the techniques used for the analysis and characterization will change. In this presentation we will discuss how HPLC/UHPLC is integrated into the workflows and how at each stage in the lifecycle process the outputs from the analyses will vary. We address how the choice of biocolumn and HPLC instrument can influence the productivity of the analysis and how it is possible to increase the productivity through method optimization. Selecting the right Liquid Chromatography to suit your needs Ian Sik Method development for fast LC analysis Bradley Skelchy When looking to increase the speed of analysis there are two possible column choices; the first approach is to reduce the particle size from the 5um and 3um columns used for HPLC to the sub 2um columns that are operated with faster flow rates for reduced analysis times. The alternative is to use a superficially porous media, Poroshell, which also provides for faster separations but tyupically operates at lower pressures. This presentation will look at the strategy for method development used to achieve fast LC analysis with these two types of columns. How to increase the lifetime of your HPLC columns and reduce system downtime Dr Linda Lloyd As we move from HPLC with 5um columns to UHPLC with sub 2um columns the requirements for maintenance increases. There are a number of practices that should be implemented to maximise the lifetime of the HPLC column and reduce the system downtime. We will discuss the importance of these practices and how they can be applied to analytical methods.
