INPENGARUH PEMBERIAN TEH HIJAU (Camellia assamica) BERBAGAI DOSIS TERHADAP KADAR FERRITIN DAN VITAMIN C PADA TIKUS

PUTIH

ABSTRAK Latar belakang : Terapi transfusi pada pasien thalassemia menyebabkan kondisi iron overload sehingga terjadi peningkatan ferritin dan penurunan kadar vitamin C tubuh akibat penggunaannya sebagai antioksidan. Teh hijau mengandung katekin yang dapat digunakan sebagai alternatif chelating agent untuk menurunkan kadar ferritin berlebih dalam tubuh. Backgorund : Transfusion therapy for thalassemia patients cause an overload iron condition that leads to increasing of ferritin level and decreasing of vitamin C level, it is caused of its function as an antioxidant. Green tea contains cathecin which can be used as a chelating agent alternative for decreasing the excess ferritin level in human body. Tujuan: Penelitian ini bertujuan untuk mengetahui pengaruh pemberian teh hijau berbagai dosis terhadap kadar ferritin dan vitamin C pada tikus putih. Objective : To find out the effect of administering green tea in various doses to the level of ferritin and vitamin C in white mice (tikus putih) Metode: Penelitian ini menggunakan metode analitik eksperimental pre and post test with control group design. Sampel 24 ekor tikus putih (Rattus norvegicus) yang telah diinduksi FeSO4 selama 30 hari dengan dosis 0,5 mg/kgBB/hari kemudian dilakukan pemeriksaan pre test kadar ferritin dan vitamin C. Sampel dibagi menjadi 6 kelompok: 1 kelompok kontrol negatif, 1 kelompok kontrol positif dan 4 kelompok perlakuan ekstrak teh hijau 1,6 gr/kgBB/hari, 2,4 gr/kgBB/hari, 3,2 gr/kgBB/hari, dan 6,4 gram/kgBB/hari selama 30 hari kemudian dilakukan pemeriksaan post test kadar ferritin dan vitamin C. Data terdistribusi normal dengan uji Saphiro-Wilk dan analisis statistika dengan uji ANOVA (Analysis of varian) dilanjutkan uji Post hoc Tukey HSD menggunakan program SPSS 17 untuk melihat perbedaan rerata pengaruh antar kelompok. Method : This study was designed as an analytic experimental pre and post test with control group. Study was conducted on 24 tikus putih (Rattus norvegicus) which has been inducted by FeSO4 as long as 30 days with a dose of 0.5 mg/Kg/day then pretest examination is conducted for measuring the level of ferritin and vitamin C. 6 groups of tikus putih (Rattus norvegicus) were formed in this study: 1 negative control group, 1 positive control group, and 4 groups was administered by green tea 1.6 gr/Kg/day, 2.4/Kg/day, 3.2 gr/Kg/day, and 6.4 gr/Kg/day as long as 30 days and after that posttest examination is conducted for measuring the level of ferritin and vitamin C. Normal data was distributed by Saphiro-Wilk test and statistical analysis by ANOVA (Analysis of varian) test then continued by Post hoc Tukey HSD which used SPSS 17 program to see the difference of average effects among groups.

Hasil: Hasil uji statistik adalah p<0,05 yang berarti terdapat perbedaan signifikan rerata antar kelompok perlakuan kadar ferritin (p=0,000) dan vitamin C (p=0,000). Result: The result of statistical test is p<0.05 which mean that there are significant difference in average of ferrtin level (p=0,000) and vitamin C level (p=0,000). Kekurangan: Kelemahan dalam penelitian ini adalah kurangnya variasi dosis dan lama pemberian teh hijau, sehingga pengaruh teh hijau mungkin belum berbeda secara signifikan. Kekurangan (Lack): This study was lacking in the variation of doses and the duration of administering green tea, so that the administration of green tea have not given any significant impact. Kesimpulan: Terdapat pengaruh dari pemberian teh hijau terhadap penurunan kadar ferritin dan peningkatan vitamin C pada tikus putih. Kadar ferritin mencapai nilai terendah pada kelompok perlakuan dengan dosis teh hijau 3,2 gram. Conclusion: There is an effect of administering green tea to the decreasing of ferritin and vitamin C level on white mice. Ferritin level reaches the lowest level on the groups with green tea dose 3.2 gram. Kata Kunci : ferritin, teh hijau, vitamin C, anti oksidan. Keywords : ferritin, green tea, vitamin c, antioxidant
