Diagnosis of PKU is made on the basis of an elevated blood Phe concentration on a repeat blood sample. The upper reference limit for Phe in whole blood or plasma in neonates is 150umol / L and slightly lower 120 in older children. A low Phe diet which reduces or normalises Phe concentrations prevents the development of the neurological and psychological changes.
